EGFR unusual mutation status in lung adenocarcinomas

چکیده
Lung cancer is the most common cause of cancer deaths in both men and women. Adenocarcinoma represents about 28% of the NSCLC cases in men and 42% in women. EGFR is a member of the ERBB family of tyrosine kinases (TK). EGFR mutations are more frequently observed in female, non-smokers, East-Asian and in patients with adenocarcinomas, and predict response to TK Inhibitors (TKIs). Sections of adenocarcinomas of the lung, formalinfixed paraffin-embedded tissues (FFPE), were selected to analyze mutations in EGFR exons 19 and 21 by DNA extraction for polymerase chain reaction (PCR). Exon 19 was studied by fragment analysis and exon 21 was studied by direct sequencing. The analysis of FISH results was done by Cappuzzo’s score to EGFR gene. Determination of EGFR protein expression was done by immunohistochemistry (IHC) (Zymed Laboratories). The author’s present two cases of lung adenocarcinoma that harbours coexisting EGFR exon 19 and 21 mutations and one case of EGFR multiple in framedeletions. The patients were female (n = 3), with mixed type adenocarcinoma overexpressing EGFR by IHC. Most reports demonstrate one EGFR mutation per adenocarcinoma. We demonstrated that a single adenocarcinoma can harbour more than one EGFR activating mutations.

Discordance of Mutation Statuses of Epidermal Growth Factor Receptor and K-ras between Primary Adenocarcinoma of Lung and Brain Metastasis
Mutations on epidermal growth factor receptor (EGFR) of adenocarcinomas of lung have been found to be associated with increased sensitivity to EGFR tyrosine kinase inhibitors and K-ras mutations may correlate with primary resistance. We aimed to explore the discordant mutation statuses of EGFR and K-ras between primary tumors and matched brain metastases in adenocarcinomas of lung. We used a se...
متن کاملDifferences in EGFR and KRAS mutation spectra in lung adenocarcinoma of never and heavy smokers
Epidermal growth factor receptor (EGFR) mutations are common in lung adenocarcinomas of never smokers, while KRAS mutations are more frequent among heavy smokers. Different clinicopathological and biological characteristics may, therefore, exist in lung adenocarcinoma according to smoking status. In the present study, a retrospective review was performed using 521 patients with surgically resec...
متن کاملEpidermal Growth Factor Receptor (EGFR) Gene Mutation Analysis in Adenocarcinoma of Lung, the First Report from Iran
Background and Objective: Epidermal growth factor receptor (EGFR) gene mutation, especially in exons 18 to 21, is an important predictor of the response rate of lung adenocarcinoma to tyrosine kinase inhibitors. There are variable reports from Asian and European countries, as well as North America, about the frequency of the EGFR mutation in lung adenocarcinoma, yet molecular s...
متن کاملThe Association of EGFR Mutations with Stage at Diagnosis in Lung Adenocarcinomas
BACKGROUND The prognostic role of epidermal growth factor receptor (EGFR) mutations in patients with lung adenocarcinomas remains controversial and the association between EGFR mutations and stage at the time of the initial diagnosis is debatable. In this study, we evaluated the association of EGFR mutations with stage at diagnosis in lung adenocarcinomas. MATERIALS AND METHODS We retrospecti...
متن کاملCorrelation of EGFR or KRAS mutation status with 18F-FDG uptake on PET-CT scan in lung adenocarcinoma
BACKGROUND 18F-fluoro-2-deoxy-glucose (18F-FDG) positron emission tomography (PET) is a functional imaging modality based on glucose metabolism. The correlation between EGFR or KRAS mutation status and the standardized uptake value (SUV) of 18F-FDG PET scanning has not been fully elucidated. METHODS Correlations between EGFR or KRAS mutation status and clinicopathological factors including SU...
